The best-case scenario for me is to play against zerg and having spotters as the first special infested as chokers, hunterlings, and kaboomers can all slow me down to some degree. I think Zagara is one of the best for speedrunning this map. Usually I wind up finishing somewhere in the middle of day 3. We finished clearing the map about 2/3rd of the way into day 2 when our armies met in the top right corner of the map.
I think the fastest time I've done on DoN was me playing Nova and my partner playing Zagara.
I'm not a speedrunner but I have played a lot. Maguro has designed some custom mutations to mimic a mini-campaign. Mutations add special modifiers to Co-op Missions, rewarding bonus experience. It is a two player cooperative mode with campaign-style objectives.
